Alfalfa is an excellent forage crop and the most widely cultivated legume forage crop in China. It enjoys the reputation of "king of forages" due to its high protein content, strong adaptability, soil improvement ability, and high economic value. Carotenoids are a class of natural fat-soluble pigments widely found in nature. They play an important role in plant photosynthesis, hormone synthesis, and color determination. They also have various biological functions, such as antioxidant, anti-tumor, immune enhancement, and visual protection. However, the molecular regulatory mechanism of carotenoid synthesis is still unclear.

Lin Hao, a researcher at the Institute of Biotechnology and the corresponding author of the paper, introduced that this study used a carotenoid synthesis-deficient Medicago truncatula mutant to successfully clone the key regulatory gene WP1 that controls carotenoid synthesis in Medicago truncatula. This gene encodes an R2R3-type MYB transcription factor, which regulates carotenoid accumulation in Medicago truncatula flowers by directly activating the expression of genes in the carotenoid synthesis pathway.

The above research systematically revealed the regulatory mechanism of plant carotenoid biosynthesis, providing an important theoretical basis for breeding carotenoid-nutrient-improved crops and high-quality functional forage varieties.

This research was funded by the Ministry of Science and Technology, the National Natural Science Foundation of China, and the Chinese Academy of Agricultural Sciences.
