        To implement the "Opinions on Accelerating the Cultivation and Development of High-Quality Manufacturing Enterprises" (鄂经信产业〔2022〕13号), and in accordance with the "Hubei Province Manufacturing Industry Single Item Champion Enterprise (Product) Cultivation Management and Recognition Methods (Trial Implementation)" and the "Notice of the Office of the Provincial Department of Economy and Information Technology on the Carrying Out of the 2022 Provincial Manufacturing Industry Single Item Champion Enterprise (Product) Cultivation Selection and Review and Evaluation" (鄂经信办函〔2022〕46号), the Provincial Department of Economy and Information Technology organized the 2022 Manufacturing Industry Single Item Champion Enterprise (Product) cultivation selection and the 2019 review of hidden champions and the work of bringing them in line with existing single-item champion enterprises (products).       Wuhan Xingchen Biology, based on the project requirements and conditions, comprehensively evaluated the company's own strength, participated in and completed the application for this project. In 2021, our company submitted the application materials according to the requirements of the high-tech enterprise application notice. After a series of expert reviews, we successfully passed the re-certification of the certification management working group and obtained the high-tech enterprise certificate issued by the government in March 2022.

科学家构建出高效异源生物合成类胡萝卜素细胞工厂

近日，《自然·通讯（Nature Communications）》在线发表了中国农业科学院深圳农业基因组研究所黄三文团队和美国麻省理工学院科研团队合作研究的成果，研究揭示了两种不同的解除酶底物抑制的代谢工程策略，构建了高效异源生物合成类胡萝卜素的细胞工厂。该研究为在细胞工厂消除底物抑制生产高价值化合物研究提供参考。   　　科研人员以解脂耶氏酵母异源生物合成β-胡萝卜素为例，报道了两种不同的解除酶底物抑制的代谢工程策略。研究首次发现番茄红素环化酶具有底物抑制效应，高浓度的底物番茄红素对该酶有强烈抑制，成为解脂耶氏酵母中类胡萝卜素生物合成的主要瓶颈。第一种策略是针对番茄红素环化酶的蛋白质工程。改良后的突变体Y27R可以完全解除底物抑制并且不降低酶活性，使得β-胡萝卜素产量显著增加且选择性达到98%。第二种策略是建立香叶基香叶基焦磷酸合酶介导的流量控制器，调控代谢流量，限制番茄红素合成速率，将其浓度控制在抑制水平之下，同时在不触发底物抑制的前提下，确保足够多的代谢流量用于目标化合物合成，获得了与第一种策略相似的β-胡萝卜素滴度和选择性。两种代谢工程策略被证明同等有效地解除了底物抑制，能够高效生产β-胡萝卜素，最终使得β-胡萝卜素的产量达到了39.5克/升。   　　该研究得到了国家重点研发计划的资助。
